Application of MALDI-TOF/MS to Analysis of Pharmaceuticals

This equipment is designed based on the matrix assisted laser desorption ionization (MALDI) and the time-of-flight mass spectrometry (TOF/MS). The equipment is capable of ionizing polymer compounds (peptide, protein, polysaccharides, synthesized polymers, etc.) and substances difficult to volatilize without decomposing them and determining their molecular weight quickly with high sensitivity.

  Principle

When laser is irradiated pulsatingly to a sample mixed with a matrix (sinapic acid, gentisic acid, etc.), the matrix absorbs the laser energy, and the sample is rapidly heated and vaporized (ionized). The generated ions are accelerated by a certain voltage and fly in a space without electric fields. The ions reach the detector in the ascending order of molecular weight. The molecular weight is determined by measuring the time of flight.

  Examples of analysis

Fig.1 An example of measurement of immune globulin IgG. The molecular ions are clearly observed in the sample having a molecular weight of approx. 140 thousand. The equipment can easily measure samples having a molecular weight of more than 100 thousand and is capable of measuring samples having a molecular weight of up to 300 thousand. Fig.1

Fig.2 An example of measurement of bovine serum albumin. In addition to molecular ions, trimer cluster ions are detected in the sample having a molecular weight of approx. 200 thousand.

Fig.5 An example of measurement of methotrexate, one of antitumor drugs. Molecular ions are clearly observed in the sample having a molecular weight of 454. The equipment is capable of measuring such low-molecular weight compounds.
